(R)-3-Amino-1,2,3,4-tetrahydrocarbazole

(R)-3-Amino-1,2,3,4-tetrahydrocarbazole is a chiral amine intermediate associated with ramatroban synthesis. It supplies the required R-configured saturated-ring center and an exocyclic amino group for sulfonamide construction. The indole nitrogen offers a second functionalization site for introducing the propionic-acid-containing substituent of the target structure.

This free amine is distinct from the S enantiomer and from unresolved aminotetrahydrocarbazole. Its existing configuration allows downstream reactions to focus on nitrogen substitution rather than establishing the stereocenter after the molecule becomes more complex. The two nitrogen sites perform different roles in synthesis: the primary amine forms the arylsulfonamide, while the indole nitrogen connects the ring system to the acid-bearing chain.
