N-(4-Aminophenyl)-N-methyl-2-(4-methylpiperazin-1-yl)acetamide

N-(4-Aminophenyl)-N,4-dimethylpiperazineacetamide is an aniline-containing intermediate used in nintedanib synthesis. The aromatic primary amine provides the reactive connection to the developing oxindole fragment, while the existing methylpiperazinyl acetamide transfers the basic side-chain portion into the final structure. The two functions are already linked through a para-substituted aromatic ring.

This intermediate is distinct from N-methylpiperazine itself: the piperazine-containing side chain and tertiary amide are already assembled. Downstream coupling therefore uses the aniline NH2 group without reconstructing the basic side chain. Its identity is also relevant when tracking residual starting material in nintedanib processes, where an unreacted aniline fragment must be distinguished from the completed coupled molecule.
