6-Hydroxy-1-tetralone

6-Hydroxy-1-tetralone is a phenolic cyclic ketone used to build substituted tetralin and dihydronaphthalene structures. The ketone enables reduction, carbon–carbon bond formation or functionalization at the adjacent carbon, while the phenolic hydroxyl supports ether formation and temporary protection. The fused-ring framework is already established in the starting material.

The selected free phenol allows the aromatic oxygen substituent to be chosen before manipulation of the ketone. Benzyl protection is one route to intermediates used in elacestrant synthesis, where the oxygen functionality is masked during subsequent steps. Alternatively, carbonyl transformations introduce new substituents on the saturated ring. These independent reaction sites support staged assembly of oxygen-substituted tetralin scaffolds.

Typical Applications

• Protected tetralone production: Used in phenolic O-benzylation to prepare benzyloxy tetralone intermediates, preserving the ketone for subsequent ring functionalization.
• Substituted tetralin synthesis: Used in carbonyl reduction or adjacent-carbon modification sequences to generate alcohol-, alkene- or carbon-substituted tetralin fragments while retaining the aromatic oxygen function.
