Piperazin-2-one

Piperazin-2-one is a cyclic amino amide used to build substituted piperazinone scaffolds. It contains a secondary amine and a lactam nitrogen with different reactivities, enabling selective side-chain attachment and staged nitrogen functionalization. The carbonyl also introduces polarity and a defined amide geometry into the six-membered ring.

The selected material is the unsubstituted piperazinone, not piperazine and not a diketopiperazine. Its free amine can participate in alkylation or acylation while the lactam remains part of the scaffold. The second nitrogen can be protected or modified in a separate operation, allowing different substitution patterns to be introduced around a preformed cyclic amino-amide framework in medicinal-chemistry synthesis.

Typical Applications

• N-Substituted piperazinone intermediates: Used in alkylation or acylation of the non-amide nitrogen to attach aromatic, heteroaromatic or aliphatic side chains to the cyclic amino-amide framework.
• Protected heterocyclic building blocks: Used to prepare selectively protected piperazinones, separating functionalization of the lactam nitrogen from later reactions at the more nucleophilic amine site.
