(S)-1,2,3,4-Tetrahydronaphthalene-1-carboxylic acid

(S)-1,2,3,4-Tetrahydronaphthalene-1-carboxylic acid is a chiral aromatic-alicyclic acid used as an acyl building block. It supplies an S-configured tetralin fragment and a carboxyl group for attachment to amines or alcohols. The partially saturated fused-ring structure introduces a rigid hydrophobic portion while preserving a defined orientation at the acid-bearing carbon.

This S enantiomer is different from the R acid and the racemate. It is relevant to synthesis of stereodefined amides, including reported dipeptidyl boronate proteasome-inhibitor intermediates. Activation of the acid allows the tetralin group to be installed late in a route, while ester formation offers a separately handled derivative for further synthetic operations. The free acid also permits formation of isolable derivatives before the final amide-coupling stage.
