Salicylamide

Salicylamide is 2-hydroxybenzamide, an aromatic amide with a phenolic hydroxyl adjacent to the carboxamide group. The phenol supports oxygen substitution, while the neighboring amide participates in carbonyl-mediated ring closure. This arrangement makes salicylamide a starting material for benzoxazinedione chemistry and for derivatives that retain the salicylamide framework.

In the selected synthetic use, salicylamide is converted into Carsalam, a cyclic protected and activated form of the hydroxybenzamide unit. That intermediate enables subsequent nitrogen alkylation in routes to salicylamidoalkanoic acids such as salcaprozic acid. Separate phenolic functionalization provides access to O-substituted benzamide derivatives. The ortho relationship of phenol and carboxamide is retained in those oxygen-functionalized derivatives.

Typical Applications

• Carsalam manufacture: Used with a carbonylating reagent in cyclization processes that form 2H-1,3-benzoxazine-2,4(3H)-dione from the adjacent phenol and amide functions.
• Salicylamidoalkanoic-acid synthesis: Used as the upstream aromatic starting material in Carsalam-based routes to salcaprozic acid and related delivery-agent structures.
• O-Substituted benzamide intermediates: Used in phenolic alkylation or acylation to prepare derivatives bearing an oxygen-linked substituent on the benzamide ring.
