Trimethyl orthobenzoate

Trimethyl orthobenzoate is a liquid orthoester reagent used in heterocycle construction and conversion of adjacent hydroxyl groups into cyclic orthoester derivatives. Its carbon bearing three methoxy groups undergoes substitution and condensation reactions, introducing a phenyl-substituted carbon unit. These reactions distinguish it from methyl benzoate, which contains an ordinary ester carbonyl.

The reagent supplies both a condensation center for nitrogen-containing ring systems and a temporary linking group in carbohydrate chemistry. Reaction with appropriately arranged hydroxyl groups can form a cyclic orthoester, while condensation with nitrogen-rich substrates generates different heterocyclic frameworks.
