Carsalam is a cyclic salicylamide derivative also named 2H-1,3-benzoxazine-2,4(3H)-dione. The ring temporarily incorporates the phenolic oxygen and amide functionality of salicylamide, while its nitrogen remains available for alkylation. This protected and activated arrangement enables attachment of a carboxyalkyl chain before release of the final hydroxybenzamide structure.
In routes to salcaprozic acid and salcaprozate sodium, Carsalam is alkylated with an omega-haloalkanoate and then opened under hydrolytic conditions. The sequence combines selective side-chain installation with recovery of the phenolic hydroxyl. Varying the haloalkyl reagent changes the attached chain while the cyclic salicylamide supplies the same aromatic hydroxybenzamide portion.
Typical Applications
- SNAC side-chain intermediate manufacture: Used in N-alkylation with an 8-halooctanoate ester to prepare the protected salicylamidooctanoate precursor.
- Related salicylamidoalkanoic acids: Used with haloalkyl reagents of other chain lengths or substitution patterns, followed by ring opening, to build related hydroxybenzamide-containing delivery-agent intermediates.
