1-(Cyclopropanecarbonyl)piperazine hydrochloride

1-(Cyclopropanecarbonyl)piperazine hydrochloride is a monoacylated piperazine salt used to introduce a preassembled cyclopropanecarbonylpiperazine fragment. One nitrogen already carries the cyclopropyl amide, while the second becomes available for coupling after neutralization. This permits construction of unsymmetrical piperazine diamides without introducing both acyl groups in the same operation.

The selected hydrochloride is an isolated feedstock form, distinct from the corresponding free amine. Its main downstream reaction occurs at the remaining piperazine nitrogen, leaving the existing cyclopropanecarbonyl group intact. In olaparib-oriented routes, this arrangement allows direct connection to the fluorinated phthalazinone-containing benzoic acid fragment. The two nitrogen positions therefore carry different, deliberately installed carbonyl substituents.

Typical Applications

• Olaparib convergent amide coupling: Used after neutralization as the amine partner for the phthalazinonylmethyl fluorobenzoic-acid fragment, joining the preinstalled cyclopropyl amide to the larger scaffold.
• Cyclopropyl-containing piperazine diamides: Used with alternative activated acids to produce unsymmetrical diamides that retain the cyclopropanecarbonyl substituent on one nitrogen.
